Decitabine

Decitabine is an analogue of the natural nucleoside 2’-deoxycytidine. It functions in the same way as 5-Azacytidine. The antineoplastic activity of this drug is dependent on its intracellular conversion to its 5'-triphosphate metabolite.

Azacitidine

Azacitidine (5-azacytidine) is a chemical analogue of the cytosine nucleoside used in DNA and RNA. Azacitidine is believed to exert its antineoplastic effects by causing hypomethylation of DNA and direct cytotoxicity on abnormal hematopoietic cells in the bone marrow. The concentration of azacitidine required for maximum inhibition of DNA methylation in vitro does not cause major suppression of DNA synthesis.

SGI-1027

SGI-1027 is a potent inhibitor of the DNA methyltransferases DNMT1 (IC50 = 0.85 µM) and DNMT3A (IC50 = 0.9 µM). It has been shown to cause the degradation of DNMT1 and to reactivate silenced tumor suppressor genes by reducing CpG island hypermethylation.
